With more than 80,000 passionate Mexican supporters creating a deafening atmosphere, Hugo Broos' side battled bravely for long periods but were ultimately undone by costly mistakes and two second-half red cards.
Backed by a passionate home crowd, Mexico threatened almost immediately. South Africa goalkeeper Ronwen Williams was called into action inside five minutes, producing a fine save to deny Raul Jimenez after the striker's powerful low effort.
However, disaster struck in the ninth minute. A defensive error while attempting to play out from the back gifted possession to Mexico, allowing Julian Quinones to pounce. The forward calmly created space before firing a low shot through Williams' legs to send the home crowd into celebration.
Mexico continued to push forward throughout the first half. Quinones remained a constant threat, while Roberto Alvarado also tested the South African defence. The hosts came close to doubling their advantage before the break when Quinones struck the base of the post after another well-worked attack.
Any hopes South Africa had of mounting a comeback after half-time suffered a major setback four minutes into the second period. Sithole was shown a red card after bringing down Brian Gutierrez as the last defender, leaving Bafana Bafana with 10 men.
Mexico took full advantage of their numerical superiority and eventually found a second goal. Raul Jimenez marked his first World Cup start with his first goal at the tournament, arriving at the back post to convert an inviting cross with a header from Alvarado.
South Africa's difficult evening worsened late on when substitute Themba Zwane received a straight red card following an altercation with Alvarado.
Mexico were also reduced to 10 men in the closing stages after Cesar Montes was sent off for a foul on Khuliso Mudau, but the result was never in doubt.
The victory gave hosts Mexico an ideal start to their World Cup campaign, while South Africa will hope to regroup quickly ahead of their remaining fixtures against Czechia and South Korea.
