Mourinho handed the former Liverpool defender a starting role at right-back after an injury to club captain Fede Valverde forced a change to his usual selection.
The decision proved successful, with Alexander-Arnold producing an outstanding display on his return to action after spending time out of the team.
Mourinho had previously suggested that Valverde could be his preferred long-term option at right-back, but the Portuguese coach was impressed by what he saw from the England international.
"Trent was really good today. He hasn't played in a while, and he comes out and drops a masterclass," Mourinho said.
"That's what happens when a team is confident and trusts each other."
The Real Madrid manager also explained that Alexander-Arnold's technical ability made him the ideal choice for the match, although he stopped short of guaranteeing him a regular starting place.
"I thought Trent, with his technical skills, was the best option for this game, but that could change in other matches," Mourinho added.
Alexander-Arnold will now hope his performance earns him further opportunities as Real Madrid prepare for a demanding week.
Los Blancos travel to Greece to face AEK Athens in the Champions League on November 4 before returning to La Liga action away against Valencia four days later.
