Jadeja was the chief destroyer, claiming 3 for 52 with his trademark accuracy and bounce, while Kuldeep Yadav chipped in with a wicket as India's spin trio kept constant pressure on the Caribbean side.
Earlier, India's day belonged to captain Shubman Gill, who struck a stylish unbeaten 129, his fifth Test hundred of the year and tenth overall. Gill played with control and flair, striking 16 fours and two sixes, delighting the packed weekend crowd.
Starting the day on 20, the 26-year-old took charge after Yashasvi Jaiswal's dismissal for 175 in the second over of the morning. The pair had already put India in a commanding position, and Gill ensured there was no let-up.
Gill added 91 runs with Nitish Kumar Reddy (43) before sharing another 102-run stand with wicketkeeper Dhruv Jurel (44). Their partnership quickened the scoring rate, forcing West Indies onto the back foot before India declared midway through the second session.
The West Indies reply began shakily when John Campbell fell for 10 after a bizarre reflex catch by debutant Sai Sudharsan at short leg.
Tagenarine Chanderpaul (34) and Alick Athanaze (41) offered brief resistance before both fell to spin, Chanderpaul edging Jadeja to slip and Athanaze undone by Kuldeep. Captain Roston Chase's duck deepened the trouble as Jadeja struck again in quick succession.
Hope and Imlach survived some nervy moments to see the day through, but India's dominance was clear. With the hosts already one up in the two-match series after an innings win in the opener, they looked well placed to seal another emphatic victory.
