The 20-year-old beat the long-standing record previously held by West Indies legend Chris Gayle, who scored an unbeaten 175 for Royal Challengers Bengaluru against the now defunct Pune Warriors in an IPL clash in 2013.
Pretorius, who last month became the youngest player to score a men's international T20 hundred with 102 against Namibia, was playing in his first game after returning to the field following a groin injury.
The left-handed batter wasted no time to get going in the Pro20 contest at the Mangaung Oval, hitting a four and six off the first two balls of the encounter before going on to reach his fifty in just 24 balls.
It took him only 16 more deliveries to get to his century, while his 150 came off 58 balls with Pretorius eventually finishing his knock with 15 fours and 13 sixes to get the Titans to an imposing total of 267-3 in 20 overs.
It was the second highest total in South Africa's domestic T20 tournament, behind the Titans' own 271-3 against the same opposition in 2022.
The Titans dismissed the home side for 143 to win the contest by a monumental 143-run margin to make it three wins from three in the tournament.
