The Royals were in early trouble when they slipped to 36-2 in the fifth over after the Asa Tribe and Lhuan-dre Pretorius lost their wickets.
The innings was resurrected when Dan Lawrence and Rubin Hermann put on a stand of 106 off 61 balls of the third wicket.
Lawrence struck his maiden SA20 fifty before he fell for a sparkling 63 from 41 deliveries, that included two fours and four sixes.
Hermann, who came into to bat at number 4, took the home side over the line in the final over with an unbeaten 65 off 45 balls.
The game came down to the final ball with the Paarl Royals needing two runs to win and Sikandar Raza deposited David Wiese over square-leg for six for wild scenes in the capacity crowd.
The Zimbabwe-born Raza finished on a vital 27 not out from 13 balls as the Royals recorded their fourth victory in seven encounters this campaign.
Earlier, visiting skipper Aiden Markram struck 66 from 46 balls as the Super Giants posted a total of 186-5 in their 20 overs after sent in to bat at the toss.
David Bedingham scored a run-a-ball 25 but the innings was given a late boost when England international bludgeoned 32 from just 10 deliveries to take the visitors over the 180-run mark.
